Pearce had been in charge of first the reserve side and, for the last four years, the first-team with the capture of the Division One West championship in season 2010/11 the stand-out achievement.
Pearce has also decided to hang up his boots from regular football at the age of 41 as he contemplates a complete rest from football.
An amicable parting will see Pearce remain a member of the club.
Cuff has been with Headington for five years and in that time has proved one of the most prolific strikers in Hellenic League football.
He will take his first training session tonight and will be in charge when Headington take on Shrivenham in the Jim Newman Cup at Clanfield on Saturday (28th July).
