Hopkins had previously been in charge of Sphinx Sports & Social and has guided them to second in the Coventry Alliance Division Two table in recent weeks.
He was on the bench for Sphinx's 3-1 victory at Tipton Town and replaces Carl Nolan, who resigned due to work commitments earlier this month.
“Luke is the ideal candidate and fits the role profile perfectly,” said club chairman Danny Cahill.
“Following the departure of Danny McSheffrey and Lee Knibbs at the end of last season it was always going to be a real challenge in replacing them partially from outside the reaches of the club.
"Luke has been within the club for many years and has a real passion for the football club.
“He has managed to pull together a tremendous team of back room staff which will assist him in what is a very time consuming and pressure role.”
Cahill also thanked Nolan and assistant Andy Taylor for the work they put in over the summer.
“Both Carl and his assistant Andy arrived in the close season and have both worked their socks off to get it right but just fell short in the end,” he added.
“It is never easy releasing managers who put a lot of time and effect with no financial gain but it became clear we needed to act sooner rather than later for everybody’s sake.
“This is not personal in any way shape or form. I have to think about the future stability of the football club and it was vital we moved quickly to resolve our current league form.”
