In a cracking tie, Fleet, managed by former Trophy winner Mark Stimson, came from behind three times to progress.
The Dockers went ahead after 10 minutes, albeit slightly against the run of play, when Tom Wood converted a low cross past Andy Walker in the home goal from close range.
Charlie Stimson pulled his dad`s side back on level terms 12 minutes later, only for the visitors to re-take the lead 2 minutes later when the Fleet defence failed to deal with a long ball and Francis Babalola lobbed the ball over Walker.
But on 36 minutes the hosts were level once more when a corner was flicked on at near post by Ross Wall past Ricky Millard in the Tilbury goal.
However, Tilbury went into half-time in front when, 4 minutes before the break, Ellis Remy`s free-kick from 25 yards beat Walker at his near post.
But the second period belonged to Thurrock, who scored twice in 5 minutes just past the hour-mark with Wall completing his hat-trick.
And although they had Loui Hazlewood – on as a half-time substitute – sent-off in the 84th minute for a second bookable offence, they held on to go through.
